The figures, for anyone assembling their own picture. Numbers worth memorising for this class: Tmax one to three days for the weekly peptides, terminal half-life about a week for semaglutide and about five days for tirzepatide, steady state at four to five half-lives, subcutaneous bioavailability high enough that site choice is irrelevant.

If somebody has the primary source to hand I would rather cite it than paraphrase it.

Following on from andrew_nyc — and this may be the naive question:

Which effects tachyphylax and which persist, because the answer explains why tolerability improves while the appetite effect keeps working?

Coming at HPLC_Greg’s question from a different direction. The version of this that has an answer is narrower than the version being asked. Narrow it and it becomes tractable; leave it broad and the thread will produce nine confident and incompatible replies.

I would rather be corrected than agreed with, if it comes to it.

Reporting back.

Closing this out: the trough rather than the peak explains the pattern I was seeing on day six, which I had been blaming on the vial.

Fair, and it stops being fair at the extremes. The middle of the range is well understood; the ends are where the honest answer is that nobody knows.
